To be ridden by her Sydney-based stable jockey Nash Rawiller, Joku will be Waterhouse's first starter at Kilmore.
A last start winner of the John Dillon Stakes, Joku must carry 59kg but is yet to finish further back than second in seven starts over the race distance of 1400m.
The Clinton McDonald-trained pair
Pacino and Rolling Stock will be worthy rivals as will this season's Moonee Valley Cup runner-up Irazu who is the only horse in the field boasting a win over the track and distance.
Others key runners are Inkster, Scenic Scene, Tongi and recent Bairnsdale Cup winner Encroacher who finished sixth in this race 12 months ago.
Last start Flemington placegetter True Tales and promising four-year-old Bertonic are also capable of winning but both have accepted for Moonee Valley on Saturday and may not take their place at Kilmore.
The Kilmore Cup is the first of five country cups to be showcased under the banner of the 2010 Melbourne Festival of Racing.
